Isolasi Alkaloid Dan Uji Aktivitas Antimalaria Dari Buah Tabernaemontanamacrocarpa Jack (Apocynaceae)

Genus of Tabernaemontana (Apocynaceae) is known as one of the rich content of alkaloids and has many pharmacological activities including an antimalarial. Tabernaemontana macrocarpa Jack is a plant that has fruit that is consideret to have a number biological activities. This research was intended to isolate the alkaloids from the fruit of T. macrocarpa and determine the chemical structure and antimalarial activity. The method in the isolation process by maceration using methanol and ultrasonication. Separation of alkaloids in the methanol extract was conducted with acid-base fractionation method then the alkaloids re-extracted after basifying with chloroform. Alkaloid contained in the total alkaloids were separated using flash chromatography and the results obtained in the form of yellowish white crystals. This compound was coded as TM1. The purity of this compound was analized by TLC and HPLC as well as determination of the melting point range. Structure of pure compound was determined using spectroscopy method including UV-vis, infrared (IR), nuclear magnetic resonance (1H, 13C, 2 dimensional). Based of spectroscopic data TM1 was similar to voaphylline or conoflorine that previously isolated from Conopharyngia longiflora. Biological activity against Plasmodium falciparum clone 3D7 in vitro showed IC50 value of 34,73 uM and it was considered as moderate antimalarial activity
