Isolasi Dna Dan Amplifikasi Dna Ikan Sengarat (Belodontichthys dinema, Bleeker 1851) Berdasarkan Gen Nd4 Mitokondrial Dari Sungai Paparan Banjir Provinsi Riau
Floodplain river ecosystems serve for the survival of fish as a habitat for fish, which are complex with rivers, creeks and lakes flood or oxbow lake. One of the fish that lives in floodplain river is Sengarat Fish (Belodontichthys dinema, Bleeker 1851). The aims of this study were to isolate DNA and amplify DNA of Sengarat Fish (B. dinema) based on Mitochondrial Gene ND4 from floodplain river in Riau. Primary ND4 was designed by fragments ND4 of Silurus asotus (GeneBank) namely ND4_F1 (forward) and ND4_ R1 (reverse) which were used to amplify the gene region of ND4 on Sengarat Fish (B. dinema) floodplain river in Riau. The results of total obtained DNA had a band which was not smear and some samples contained a thin band. The DNA resulted from PCR amplification had more than one band, due to the primers used were not specific for the Sengarat Fish (B. dinema).
