Isolasi Enzim Bromelin Dari Bonggol Nanas (Ananascomosus(L) Merr)
Bromelien is one of the sulfihydryl protease enzymes obtained through the isolation process of bromelien enzymes capable of hydrolyzing the peptide bonds of proteins or polypeptides into smaller molecules of amino acids. This study aims to extract the enzyme bromelien from pineapple cobs and determine the activity of bromelien enzyme. The crude extract of bromelien enzyme was obtained by precipitation method by salt with different saturation level. In this study, the pineapple cobs were extracted and the crude extract was obtained, then precipitation was performed using ammonium sulfate ((NH4)2SO4) salt and on the final result will be determined the unit activity using the casein substrate by Kunitz method modified and the method used for analysis of enzyme activity is spectrophotometric method using spectrophotometer. Precipitation was performed by addition of ammonium sulfate ((NH4)2SO4) with variations of 40%, 50%, and 60%, and enzyme dialysis process using nitrocellulose membranes. The result of enzyme isolation on pineapple cobs was obtained from centrifugation process with yield of bromelien enzyme on 40% ammonium sulphate salt; 50%; 60% were 14.22%, respectively; 18.95%; 24.84%. Bromelien enzyme that has been tested its activity has value of enzyme activity with 40% ammonium sulphate salt; 50%; and 60% were 0.04 U/ml, respectively; 0.08 U/ml; and 0.15 U/ml. This difference in the activity of isolated enzymes may be due to differences in fruit maturity, temperature, and pH levels.
