Isolasi Senyawa Terpenoid Ekstrak Metanol Tanaman Bangun-Bangun (Plectranthus Amboinicus ) Serta Uji Aktivitas Antibakteri Dan Antioksidan
Plectranthus amboinicus (Lour.) is a perennial plant belonging to the family Lamiaceae which have widely used in traditional herbal medicine. In the present study, the extracts of the leaves of P. amboinicus by different solvent were evaluated for their antioxidant and antibacterial properties. The fresh leaves of the plant were extracted using methanol subsequently fractionated using n-hexane and dichloromethane. The antioxidant activity was determined by DPPH free radical scavenging assay and the antimicrobial activity was tested against Escherichia coli and Staphylococcus aureus by agar well diffusion assay. The antioxidant activity test results showed that dichloromethane extract of the leaves had a highest antioxidant activity with an IC50 value of 50.2 μg/mL however methanol and n-hexane extracts were active in DPPH with IC50 value of 71.3 μg/mL and 205.9 μg/mL, respectively. Moreover, the dichloromethane and methanol extracts have shown a promising antibacterial activity in E. coli. Furthermore, two compound PA-Hx-1 and PA-Hx-2 were isolated from n-hexane and one compound was isolated from dichloromethane of P. amboinicus leave. They compound have been identified 6-acetyl-7-hydroxyroyleanone, 6-7-dehydroxyroyleanone and stigmasterol. Their structures were elucidated by extensive spectroscopic methods including 1D and 2D NMR experiments as well as HR-MS. The diterpine compounds were reported in previously study isolated from P. bally and P. grandidentatus but these compounds have never been reported from P. amboinicus. The findings indicated the potentiality of P. amboinicus for further study as herbal medicine.
