Isolasi Metabolit Sekunder Dan Uji Antioksidan Ekstrak Etanol Daun Bakau Minyak (Rhizophora Apiculata)
The plant Rhizophora apiculata, also known as mangrove oil is one of the mangrove species from Rhizophora genus, and Rhizophoraceae family. This plant contain many active compounds that have antibacterial, antifungal, antitumor and antioxidant activities. Preliminary tests showed secondary metabolites of flavonoids, phenolics and terpenoids. Isolation of secondary metabolites from leaves of Rhizophora apiculata begins with maceration using ethanol which is partitioned with hexane and ethyl acetate solvents. Ethanol extract was separated by VLC chromatography using gradient solvents and produced 5 fractions which were then re-separated by chromatographic columns so that the crystals coded with IMS-1 were 36 mg. Crystal IMS-1 was further refined using flash chromatography and produced pure greyish-green compounds which were coded IMS-2 as much as 4 mg. Characterization of IMS-2 compounds using UV spectroscopy showed maximum absorption at wavelengths 206, 259, 303 and 362 nm while characterization with FT-IR was obtained at wave numbers 3423, 1596, 1502 and 2939 cm-1. Pure IMS-2 compounds are thought to be flavonoid compounds. IMS-1 compounds are antioxidants which are quite high with an IC50 value of 15.2 µg/mL as measured by the DPPH method.
