Analisis Sekuens Barkode Dna Pada Daerah Ndhc_Trnv Intergenic Spacer Pada Tumbuhan Cocor Bebek
ABSTRACT
DNA barcode is a short sequence that can be used in DNA barcoding. One of the
DNA barcodes in plants is the ndhC_trnV intergenic spacer sequence. The
purpose of this study was to analyze and compare the DNA barcode sequences in
the ndhC_trnV intergenic spacer in two species of cocor bebek A and B in
Pekanbaru, Riau. The research steps consisted of morphological observations,
sample preparation, total DNA isolation, total DNA electrophoresis, PCR, PCR
product electrophoresis, sequencing and data analysis. Morphological
observations showed that the species name of cocor bebek A can only be
determined up to the genus level, namely Kalanchoe sp., While cocor bebek B can
be determined to the species level, namely K. x laetivirens. The DNA sequences
from ndhC_trnV intergenic spacer in Kalanchoe sp. successfully obtained with a
size of 949 bp and in K. x laetivirens 954 bp in size. The DNA sequences of
ndhC_trnV intergenic spacer in both sampels had the highest similarity with K.
daigremontiana respectively 98,85% and 99,90%. Based on this sequence, ten
different nucleotides were found between Kalanchoe sp. and K. x laetivirens, i.e.
nucleotide numbers 81, 250, 327, 477, 479,480, 481, 482, 483 and 526.
