Analisis Filogenetik Spesies Salacca Di Daerah Aceh Berdasarkan Sekuen Gen Rbcl
ABSTRACK
Aceh is one of the producing areas of salak cultivation type and wild type because it has a unique topography. Deforestation and land clearing in Aceh resulted in reduced salak habitat and salak wealth in Aceh. The lack of information about salak kinship in Aceh and taxonomic studies based on morphological character are difficult to do in salak, so a molecular approach is needed to facilitate the study of taxonomy. This study aims to analyze and reconstruct the kinship relationship of Salacca species in Aceh based on rbcL gene. Extraction using the CTAB method with modifications. DNA sequencing was conducted at the First Base Laboratorium Malaysia. Phylogenetic analysis using Maximum Parsimony (MP) and Neighbor Joining (NJ) methods in PAUP* application version 4.0a169. The results of the analysis using the Maximum Parsimony method showed that there are two clades in the group, consisting of the clad I Salacca sp. (NH.07) Lembah Seulawah and Salacca sp. (Jgr) Kutacane and clad II are composed of Salacca sp. (L1) Leuser. Based on the matrix value of genetic distance from the method Neighbor Joining, obtained information that Salacca sp. (NH.07) Lembah Seulawah is more primitive than the other two salak samples due to long genetic distances.
