Keanekaragaman Genetik Jarak Kepyar (Ricinus Communis L.) Di Kepulauan Meranti Berdasarkan Karakter Vegetatif Dan Random Amplified Polymorphic Dna (Rapd)
Castor plants (Ricinus communi L.) produce non-edible castor oil that can be used
as material raw in a various industries such as cosmetics, pharmaceutical, and bio
fuel. It has been reported that this species is able to survive either in the marginal
area or acidic soil. Therefore, castor plants are possibly suitable as an agroforestry
commodity in peat land area. To support castor-based agroforestry, we explored
and analyzed genetic diversity of castor germplasm grown in Kepulauan Meranti
based on vegetative morphological characters and RAPD bands profile. The study
will facilitate castor breeding program in order to provide castor cultivar adaptable
to peat soil. A total of 11 castor plants were sampled in agroforestry area managed
by KPH (Kesatuan Pengelolaan Hutan) Tebing Tinggi and PT. Meranti Energi
Alam. We isolated genomic DNA from young fresh leaves and amplified using
four RAPD primers. Morphological and moleculer data were analyzed using
NTSYSpc 2.02. The results showed that the samples phenotypes varied in leave
and stem organs with the similarity ranged 0.35-0.95. All RAPD primers
generated polymorphic bands with 70.45% polymorphism. In addition, the
average of PIC value was 0.21 with the highest PIC (0.34) was provided by OPA-
04 primer. Based on RAPD profiles, similarity amongst samples was 0.51-0.94
and grouped all samples at the level of 66%. This results indicated that genetic
diversity of the population was categorized as low to medium. We suggested that
castor germplasm in Kepulauan Meranti may contribute to castor genetic
improvement program.
