Cair Dengan Aktivitas Litik Terhadap E.Coli Isolasi Bakteriofag Dari Limbah
Bacteriophages or phages are viruses that infect certain specific bacteria. The use
of phages has begun to be applied in several fields, one of which is in the medical
field. Therapy using phages is used to treat or prevent bacterial infections,
especially in multi-drug resistant (MDR) bacteria which is the main indication for
the use of phage therapy in some cases. MDR Escherichia coli is one of the bacteria
that has an MDR strain which has become a health problem in several countries.
E.coli is a gram-negative bacillus that is the organism that causes many diseases
such as diarrhea, dysentery, uncomplicated cystitis, pneumonia, bacteremia, and
peritonitis. In this study, a lytic phage that can infect E.coli was isolated from
wastewater from a hospital sewage treatment plant. This study aims to identify
phages and see their antibacterial ability against E.coli using the spot test method.
This study also aims to determine the concentration of phage stock, and to find the
Minimum Bactericidal Concentration (MBC) of the stock.. This research is a
descriptive laboratory study that was tested on spot test and plaque assay methods.
Phage stock of phage#1a which is the final product in this study showed positive
spot test results and had a concentration of ±7.7x106 PFU/mL. The results of this
study indicate that phages can be isolated from hospital wastewater and have been
tested to be used as antibacterial agents. It is hoped that the isolated phage can be
used to overcome the problem of antibiotic resistance which is increasing every
year.
