Pertumbuhan Pediococcus Pentosaceus Strain 2397 Pada Berbagai Sumber Nutrisi Dan Aktivitas Antimikroba Terhadap Staphylococcus Aureus
ABSTRACT
The addition of nutrient sources could increase lactic acid bacteria growth and antimicrobial activity against pathogenic bacteria. Pediococcus pentosaceus strain 2397 produced bacteriocin with a molecular weight of 14.4 kDa. The purpose of this study was to determine the best carbon and nitrogen sources that increase the growth and antimicrobial activity of Pediococcus pentosaceus strain 2397 against Staphylococcus aureus. This research was conducted experimentally using a completely randomized design with ten treatments and four replications. The treatments in this study were S0 = without the addition of carbon and nitrogen sources, various sources of carbon (S1 = addition of 2% glucose, S2 = addition of 2% fructose, S3 = addition of 2% maltose, S4 = addition of 2% lactose and S5 = addition of 2% sucrose) and nitrogen nutrients (S6 = addition of 2% peptone, S7 = addition of 2% beef extract, S8 = addition of 2% yeast extract and S9 = addition of 2% ammonium sulfate), each with a concentration of 2%. The results showed that the source of 2% carbon or nitrogen nutrients significantly affected the growth of Pediococcus pentosaceus strain 2397 and antimicrobial activities of supernatant cell-free and crude bacteriocin against Staphylococcus aureus. The S7 treatment had the largest absorbance value of 2.72 nm. In according to, the S6 treatment (addition of 2% peptone) the largest cell-free supernatant clear zone was 20.33 mm. The largest crude bacteriocin clear zone was obtained in S8 treatment using 2% fructose with an inhibition zone diameter of 23.17 mm.
