Pertumbuhan Pediococcus Pentosaceus Strain 2397 Pada Berbagai Sumber Nutrisi Dan Aktivitas Antimikroba Terhadap Listeria Monocytogenes
ABSTRACT
The additional nutrient sources could increase the growth and antimicrobial activity of lactic acid bacteria (LAB) against pathogenic and spoilage bacteria by producing antimicrobial compounds. Pediococcus pentosaceus strain 2397 is one of the LAB that can produce bacteriocin as an antimicrobial compound. The purpose of this study was to evaluate the growth of Pediococcus pentosaceus strain 2397 on various nutritional sources and antimicrobial activity against pathogenic bacteria Listeria monocytogenes. This research was conducted experimentally using a completely randomized design with ten treatments and four replications. The treatments in this study were various sources of carbon and nitrogen nutrients (S1 without additional carbon and nitrogen sources, S2 2% lactose additional, S3 2% maltose additional, S4 2% sucrose additional, S5 2% glucose additional, S6 2% fructose additional, S7 2% peptone additional, S8 2% yeast extract additional, S9 2% beef extract additional and S10 2% ammonium sulfate additional). The data obtained were analyzed statistically using analysis of variance and continued with Duncan's multiple range test at the 5% level. The results showed that the source of carbon and nitrogen nutrients 2% significantly affected the growth of Pediococcus pentosaceus strain 2397 as well as antimicrobial activities of supernatant cell-free and crude bacteriocin from strain 2397 against Listeria monocytogenes. The S9 treatment (2% beef extract additional) had the greatest absorbance value of 2.72 nm, the S4 treatment (2% sucrose additional) had the largest cell-free supernatant clear zone of 12.33 mm and the S9 treatment had the largest crude bacteriocin clear zone of 16.10 mm (with the additional of 2% beef extract).
