SUMMARY
Depression is a mental disorder that can reduce quality of life, cause disability and death. Antidepressants commonly used to treat depression still has side effects, so alternative antidepressants such as herbs that has fewer side effects were needed. Old areca nut seeds has been investigated for their antidepressant activity based on in vivo test using the forced swim test (FST) method on animal experiment. The aim of this study was to determine the antidepressant activity of the ethyl acetate extract of old areca nut seeds by simultaneously using the FST test in order to identify the effect of the demonstrator on the test mice. This study started with the extraction of old areca nut seeds using ethyl acetate solvent and continued with phytochemical tests. The mice to be tested were first injected with different treatments, namely 0.9% saline, 10, 50 and 100 mg/kg ethyl acetate extract of old areca nut seeds, and fluoxetine 20 mg/kg, which was performed 1 hour before acute FST. The best obtained extract dose was then used for 7 consecutive days and a sub-chronic FST test was performed on the 7th day. Toxicity test was performed after sub-chronic FST. Ethyl acetate extract of old areca nut seeds has antidepressant effect based on the percentage reduction in immobility time of double and single test animals which were not significantly different (17.77% and 17.29%, sig (2-tailed) > 0.05). Ethyl acetate extract of old areca nut seeds given for 7 consecutive days had no toxic effect on the mice because there was no accumulation of yellow fluid, inflammation in the gut and no tumor-like tissue growing in the mice’s livers.
