Evaluasi Keamanan Probiotik dan Aktivitas Antioksidan Lactobacillus Fermentum Inacc B1295 yang Dienkapsulasi Cellulose Microfiber dari Daun Kelapa Sawit Selama Penyimpanan
Probiotics are living microorganisms that are capable of providing beneficial effects on health when consumed in sufficient quantities. This study aimed to evaluate the safety and antioxidant activity of Lactobacillus fermentum InaCC B1295 encapsulated by cellulose microfiber (CMF) from oil palm leaves (OPL) at different temperatures and storage times. This research was carried out experimentally, consisting of descriptive analysis of safety evaluation and statistical analysis of antioxidant activity using a factorial completely randomized design (CRD). The experimental unit consisted of two factors, namely storage temperature (room and cold temperature) and storage time (0, 14, and 28 days). This study included tests for hemolytic activity, gelatinase, and cytolysin synthesis as part of its safety evaluation. It also included tests for antioxidant activity, including hydroxyl radical scavenging (HRS) activity and DPPH radical scavenging ability. Data obtained from the safety evaluation were analyzed descriptively and antioxidant activity data were analyzed statistically using analysis of variance (ANOVA), then analyzed with duncan's multiple range test (DMRT) at 5% level. The results showed that during storage at different temperatures Lactobacillus fermentum InaCC B1295 encapsulated in CMF OPL was able to maintain safety properties with negative results in all three safety evaluation tests. The results of variance showed that storage temperature, storage time, and their interactions significantly affected (P˂0.05) DPPH radical scavenging activities ranging between 36,94‒251,01 ppm, and HRS ranging between 53,72‒69,41%. DPPH and HRS radical scavenging activities decreased with storage time. The antioxidant activity of DPPH and HRS was higher at cold storage than at room temperature.
