Pemanfaatan Ekstrak Tomat Dan Lemon Sebagai Bahan Anti Browning Pada Tahap Inisiasi Eksplan Pisang Raja (Musa Paradisiaca Var. Raja) Secara In Vitro Sebagai Rancangan Handout Materi Inovasi Teknologi Biologi Sma
Bananas are one of the horticultural commodities from the fruit group that is very popular with the community and has a fairly high amount of production. In the banana propagation process, the problem that occurs is browning so that compounds that can minimize browning are needed by using antioxidant compounds, including the use of ascorbic acid sourced from natural ingredients, namely tomato and lemon extracts. The results of this research will later be used as a handout to be used as a teaching material in high school Biology learning in phase E in class X.
This research was conducted at the Tissue Culture Biotechnology Laboratory at the Coffee and Cocoa Research Center, East Java This research is an experimental study using a Complete Random Design (RAL) with two treatments. This study included 8 treatments, each of which was repeated 5 times. Each test includes 1 petri planted with 3 explants. The parameters measured were the percentage of live explant, browning intensity, percentage of calluses appearing, percentage of embryos appearing, explant color and explant growth. Then the results of the observations were analyzed using quantitative descriptive analysis and a further test of the Duncans Multiple Range Test (DMRT) was carried out at the level of 5%.
Based on the study, it was concluded that the concentrations that function as anti-browning in Plantain explants are S2 (Tomato Extract 100 ml/L) and S5 (Lemon Extract 100 ml/L) treatments. The concentrations that function in explant regeneration are S2 treatment (Tomato Extract 100 ml/L) and S0 treatment (Control). Based on the results of the research data, it has the potential to be one of the learning resources in the form of handouts that can help students in learning on the concept of biological technology innovation, especially in the field of tissue culture for high school grade X students.
