Biosintesis Asam Amino Dan Aktivitas Antioksidan Pediococcus Acidilactici Inacc B310 Menggunakan Substrat Glukosa Dari Limbah Batang Kelapa Sawit
Amino acids and antioxidants were compounds that can be synthesized through the metabolism of microorganisms. One of the microorganisms that can produce these two metabolites is Pediococcus acidilactici. The purpose of this research was to determine the interaction between the concentration of Pediococcus acidilactici InaCC B310 and the length of incubation time used in amino acid biosynthesis and antioxidant activity. This study was carried out experimentally in a factorial complete randomized design. The experimental unit consisted of two factors, namely incubation time (24 and 48 hours) and bacteria concentration (1, 3, and 5%) with three replications each to obtain 18 experimental units. The parameters observed in this study were LAB growth, amino acid levels (alanine, glutamic acid, glycine, and tyrosine levels), and antioxidant activity 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging. The LAB growth data and amino acid levels (alanine, glutamic acid, glycine, and tyrosine) obtained were analyzed statistically by analysis of variance (ANOVA) and continued with the duncan multiple range test at 5% level. Meanwhile the antioxidant activity data obtained were analyzed descriptively. The results of the analysis of variance of LAB growth showed that incubation time, bacterial concentration, and their interaction had a significant effect (P < 0.05) with values ranging from 2.068 to 2.843. The results of the analysis of variance of alanine, glutamic acid, glycine, and tyrosine levels showed that the incubation time and its interaction had no significant effect, but the treatment of bacterial concentration had a significant effect, alanine is the highest with values ranging from 4.174–4.644 µg/L. The observation of antioxidant activity showed that the antioxidant activity produced is included in the moderate to weak group with IC50 values ranging from 131.662–1.186,233 ppm.
