Seleksi Dan Uji Antagonisme Isolat Jamur Endofit Tanaman Manggis (Garcinia Mangostana L.) Terhadap Phytophthora Palmivora Penyebab Penyakit Busuk Pangkal Batang Pada Tanaman Jeruk Siam Serta Identifikasinya
ABSTRACT
Mangosteen plant (Garcinia mangostana L.) is one of fruticulture plants in horticultural plant groups which is mostly rare infected by plant diseases. The data on disease incidence and intensity have been rarely reported so far. This is assumed that the plant has natural resitance systems derived from secondary metabolites compounds such as triterpene, mangostine, tanein, resin, antosianin, and xanthone and the presence of endophytic fungi in the plant tissues that can control the diseases, such as root rot by Phytium sp. and Phytophthora sp. Endophytic fungi have been reported to have a broad disease control system potency as biological control agents due to their antagonism mechanism activities. This study aims to select isolates of endophytic mangosteen fungi that have a high antagonism activities to P. palmivora which causes basal stem rot disease on Siamese citrus and to identify them to the genus level. The research methods used in this study were exploration, observation and experimentation. The isolation results found 8 isolates of endophytic fungi that were all hypovirulent, and 3 isolates have higher inhibition ability, the biggest colony diameter and the fastest growth rate, respectively. Isolate E6 showed the highest inhibition ability (70.66%). The types of hyperparasitism interactions are lysis (E5 and E6) and entanglement (E4). Identification shows that the isolate E4 belongs to the genus of Pestalotia and isolates E5 and E6 belong to the genus of Trichoderma. Those isolates have a higher antagonism activities due to their fastest growth rate, antibiosis, higher inhibition ability, and mycoparasitism action and so that potential to become a biological agency to control P. palmivora on of Siamese citrus plants.
