Isolasi Senyawa Metabolit Sekunder Dan Uji Aktivitas Antibakteri Dari Ekstrak Metanol Kulit Batang Pulai Basung (Alstonia Spatulata)

Alstonia is a genus of plants from the Apocynaceae family plants that dwell in tropical and subtropical regions. This plant has been widely used by the community as a traditional medicine, namely to treat various conditions, such as malaria, diarrhea, fever, dysentery and diabetes. Several studies have succeeded in isolating secondary metabolite compounds from Alstonia and have tested their bioactivities such as antibacterial, cytotoxic, anti-inflammatory, antioxidant and antifungal. The aim of this study was to isolate secondary metabolite compounds from A. spatulata and evaluate the in silico antibacterial bioactivity. The bark of this plant was extracted using methanol. The concentrated methanol extract was separated by flash chromatography method to obtain pure compounds. The results of flash chromatography obtained 33 vials. These were grouped into 11 fractions based on their TLC profiles. Fraction 2 produced a yellowish white solid. Then the isolated compound was tested for purity by thin layer chromatography and melting point test. Characterization of the compound structure was analyzed using FTIR and NMR spectroscopy. Based on the results of structural elucidation obtained a mixture of 2 compounds, namely compounds 24-AS-FII-1, and 24-AS-FII-2 respectively identified as stigmasterol acetate and β-sitosterol acetate. Both compounds were tested in silico (molecular docking) using PBP receptors (PDB: 3HUN, 6G9S) and DNA gyrase (PDB: 3U2D, 4ZVI), the compounds showed strong affinity to 4 receptors, according to molecular docking results. The conclusion of this study shows that the isolated compounds of A. spatulata can be utilized as antibacterial agents.
