Analisis Sekuens Barkode Dna Pada Daerah Atpb-Rbcl Intergenic Spacer Pada Tumbuhan Pandan (Benstonea Sp.) Asal Riau
Benstonea sp. origin from Riau is one of plants that are often found in Kajuik Lake. The adaptation pattern possessed by Benstonea sp. is very unique. Benstonea sp. has the ability to live in swamp flooding areas that are often inundated. However, these plants are still able to live and adapt well in their environment. The species name of this Benstonea sp. not yet be identified. DNA barcoding is a technique designed to make molecular identification quickly and accurately based on a short-sized DNA sequence from a particular gene or region of genome such as atpB-rbcL integenic spacer. Morphological and molecular analysis using some DNA barcodes such as matk, rbcl, trnL-trnF intergenic spacer still cannot determine the species name of the Benstonea sp. from Riau so that a study was conducted for identification of the Pandan plant (Benstonea sp.) based on the sequence of the atpB-rbcL intergenic spacer. Research procedures included sampling, total DNA extraction, electrophoresis, PCR, sequencing and data analysis. Data analysis was performed using BLASTn program, BioEdit version 7.0. and MEGA version 6.0. In this study, atpB-rbcL integenic spacer fragments were obtained measuring around 851 bp. The sequence had 97.28%-97.79% similarity to the other atpB-rbcL intergenic spacer of several plants. Because no one has a 100.00% similarity to Benstonea sp. thus the species name of Benstonea sp. still cannot be determined yet and this plant is possible a new species.
