Uji Degradasi Zat Warna Reactive Black 5 Oleh Trichoderma ASPERELLUM Lbkurcc1
SUMMARY
Reactive black 5 (RB5) is a synthetic azo type dye that is widely used in the textile industry for dyeing fabrics. One method that can be used to degrade synthetic dyes such as RB5 is to use biological methods, namely directly using live fungal cells or using laccase enzymes. Trichoderma asperellum is a fungus that can produce laccase enzymes. T. asperellum LBKURCC1 is a fungus isolated from plantation soil in Riau, Indonesia, known to produce laccase. To be able to determine the ability of T. asperellum LBKURCC1 live cultures and laccase to remove RB5 dye, several RB5 dye removal tests were carried out, using live culture cells and semi purified laccase of T. asperellum LBKURCC1. Dye removal test included measurement of dye bioremediation, biosorption and biodegradation. Incubation of 50 ppm RB5 with live cultures of T. asperellum LBKURCC1 at room temperature (30oC, pH 6,5) for 24 hours resulted in 22% bioremediation, 3,2 % biosorption and 19,1% biodegradation of the RB5 dye. Biodegradation of RB5 by T. asperellum LBKURCC1 live cultures indicated that RB5 was degraded by extracellular enzymes produced by the filamentous fungi. To test if this biodegradation was due to laccase action on RB5, biodegradation tests were carried out in which RB5 was incubated with semi purified laccase. The final laccase activity used for degradation test of 50 ppm RB5 at pH 6,5 was 1,70 U/L. From the RB5 dye biodegradation test using the 20-60% ammonium sulfate laccase enzyme, it was found that the 20-60% ammonium sulfate laccase enzyme was unable to degrade RB5 dyes for 5 days which was marked by an increase in absorbance until the 5 days above 100%.
