Biosintesis Asam Amino Dan Aktivitas Antioksidan Enterococcus Faecalis Inacc B134 Menggunakan Substrat Glukosa Limbah Batang Kelapa Sawit
ABSTRACT
Indonesia is known as an agricultural country that relies heavily on the agricultural sector, one of which is oil palm plantations. Oil palm plants produce oil palm trunk waste which can be used as a substrate for the growth of lactic acid bacteria (LAB). One of the LAB strains that can produce amino acids and antioxidants is Enterococcus faecalis. The purpose of the research was to determine the interaction between the concentration of Enterococcus faecalis InaCC B134 and the length of incubation time on the biosynthesis of amino acids and antioxidants using glucose as a substrate from oil palm stem waste. The study was carried out experimentally in a factorial complete randomized design. The experimental unit consisted of two factors, namely incubation time (24 and 48 hours) and bacteria concentration (1, 3, and 5%) with three replications each to obtain 18 experimental units. The parameters observed in this study were LAB growth, amino acid levels (alanine, glutamic acid, glycine, and tyrosine levels), the data obtained were analyzed using analysis of variance and continued with the duncan multiple range test at 5% level. The antioxidant activity observation data were conducted using descriptive qualitative tests. The research results also showed that the absorbance value for LAB growth ranged from 1.923 to 2.768. The highest amino acid produced was alanine ranging from 4,196‒4,527 μg/L which increased as the concentration increased. The descriptive test results showed that E. faecalis InaCC B134 which used growth media from palm oil stem waste glucose had qualitative differences in the antioxidant activity values. The antioxidant activity value produced ranged from 197.78–596.77 ppm (which is included in the weak to very weak group).
