Analisis Empat Barkode Dna Pada Famili Fabaceae

DNA barcodes are short DNA sequences used in molecular identification of organisms. The Fabaceae family is a member of the Fabales order characterized by pod-type fruits. This study used 30 species (consisting of 6 genera, each genus has 5 species). This study aims to analyze four DNA barcodes (rbcL, matK, trnL-trnF IGS and ITS) in the fabaceae family. This study used unilocus and multilocus in DNA phylogenetic analysis. Unilocus and multilocus procedures include collection, alignment, cutting, and analysis of DNA sequences, only in multilocus there is a merging of DNA sequences after cutting the DNA sequence. Unilocus results showed that the ITS barcode had the highest level of variability, substitution rate, and diversity, while rbcL had the highest conservative site. The analysis of the four unilocus was able to separate different genera into separate groups and multilocus showed that certain barcode combinations, such as ITS+matK and matK+rbcL were able to separate different genera into separate groups. The four DNA barcodes (rbcL, matK, trnL-trnF IGS and ITS) can be used in phylogenetic analysis of the Fabaceae family. Barcode combinations that can be used in phylogenetic analysis in the Fabaceae family include ITS+matK, matK+rbcL, ITS+trnL-trnF IGS+matK, ITS+matK+rbcL and ITS+trnL-trnF IGS+matK+rbcL.
