Analisis Sekuens Nbs-Lrr (Nucleotide Binding Site-Leucine Rich Repeat) Pada Pisang Awak Dan Pisang Lilin
Fusarium oxyforum f. sp cubense (Foc TR4) is agent of Fusarium wilt disease that significantly threaten banana existence. Two banana cultivars, awak and lilin, have been previously reported potential resistant against Fusarium wilt disease. NBS- LRR (Nucleotide Binding Site-Leucine Rich Repeat) gene is the largest group of R genes that play an important role in disease resistance. Purpose of this study was to analyze the NBS-LRR sequences of awak and lilin banana. In this study, total genomic DNA was extracted from young leaf tissues. Electrophoresis result from the two samples showed an amplicons size of 500 bp using NBS F and NBS R primer pair. Subseguently, the PCR product of two banana samples were sequenced. In addition, nucleotide bases were converted into deduced amino acid sequences. The result of this study showed that deduced amino acid sequences of pisang awak and pisang lilin contained NB-ARC domains and P-loop motif. The sequences obtained in this study have the potential as a resistant genetic markers in banana.
