Identifikasi Jamur Selulolitik Indigenous Pada Kawasan Mangrove Desa Mengkapan, Kecamatan Sungai Apit, Kabupaten Siak
Abstract
This research will be conducted from February-June 2023. Mangrove litter and sediment sampling was carried out in the mangrove ecosystem of Mengkapan, Sungai Apit District, Siak Regency. The purpose of this study is to identify the genus of indigenous cellulolytic fungi found in the mangrove ecosystem of Mengkapan. The methods used in this study are survey methods and experimental methods. The procedure in this study begins with determining the location of sampling and measuring the physical parameters of waters and the chemistry of total sediment organic matter. Furthermore, samples of mangrove litter and sediment were taken and prepared to isolate cellulolytic fungi. After isolation, macroscopic and microscopic characterization was carried out for the identification of cellulolytic fungi. Then the cellulolytic fungal cellulase enzyme activity test was carried out by adding a 0.1% congo red indicator and observed the resulting clear zone activity, and the calculation of the fungal cellulolytic index was carried out. The cellulolytic fungi isolated were as many as 5 isolates, namely 4 genera Trichoderma and 1 genus Aspergillus. Based on the results of tests that have been carried out, all genera found in this study are able to produce cellulase activity as evidenced by the appearance of clear zones in each isolate. It is known that Trichoderma fungi with SRA isolate code have a cellulolytic index value of 1.34 which includes medium criteria and the genus Aspergillus has a cellulolytic index value of 0.94 which includes low criteria. Based on degradation tests that have been carried out, it is known that Trichoderma and Aspergillus fungi are able to degrade litter as evidenced by significant changes in banyan leaf litter.
