Pengaruh Pemberian Hormon Iaa Dan Bap Terhadap Pertumbuhan Tunas Anggrek Dendrobium Phalaenopsis Fitzg Secara In Vitro
This research aims to determine the effect by using IAA and BAP hormones as well as to get a combination of IAA and BAP hormones for the best shoot growth of explants D. phalaenopsis Fitzg orchid with in vitro fertilisation. The research has been carried out over three months starting from May to August 2012. The method used is an experimental method using experimental design completely randomized design (CRD) factorial with 16 treatments with two factors including the concentration of Vacin and Went (VW) media combined with IAA concentrations of 0, 0,5, 1, 1,5 ppm and BAP concentration 0, 1, 2, 3 ppm with 3 replications. The procedure is the preparation of planting materials research, sterilization equipment, provision of VW stock solution media, explants tissue culture, explant maintenance, observation and analysis of the data using variant analysis (ANAVA) and Duncan's Multiple Range Test (DMRT) with level of trust 5%. Parameters measured were percentage of live explants, while shoots emerging, number of shoot, explants height and number of root. The results showed while shoots emerging parameters is significant effect on single hormone of IAA and BAP hormones, while the number of shoots parameters showed only a single treatment of IAA hormone are significant, then the percentage of live explants parameter, the explants hight, the number of roots was not significant. Based on the results of this study concluded that the best combination of IAA and BAP hormones to produce the fastest emerging shoots we can use I0B3 treatment, to produce the highest number of shoots use I1,5B1 treatment, to produce the highest explants use I1B1 treatment and to produce the highest number of roots use I0,5B1 and I1,5B0 treatments. But overall the best treatment is I1,5B1.
