The current textile waste treatment system uses physico-chemical method
that requires further processing. For this reason, it is necessary to apply a
bioremediation method using live cultures of T. asperellum LBKURCC1 because
Trichoderma asperellum LBKURCC1 is a fungal strain isolated from the
rhizosphere of cocoa in Riau, Indonesia, where mycelia from the fungus T.
asperellum LBKURCC1 is able to produce laccase with an optimum activity of
0.072 U/mL. This study aims to determine the ability of bioremediation,
biosorption and biodegradation of the mycelia of the fungus T. asperellum
LBKURCC1 as well as degradation using crude laccase extract against Azo dye,
Basic Blue 66 (BB66) and Anthraquinone dye, Acid Blue 25 (AB25) with and
without mediator 1-hydroxybenzotriazole hydrate (HBT).
The results showed that the bioremediation, biosorption and
biodegradation of live culture T. asperellum LBKURCC1 against BB66 dye was
found in at pH 5.5 and 6.5. Bioremediation of BB66 dye at pH 5.5 was (8.90 ±
1.6)%, with contribution of (6.81 ± 1.23)% due to biosorption and (2.09 ± 0.3)%
due to enzymatic biodegradation. Bioremediation of BB66 dye at pH 6.5 was
(3.89 ± 0.1)%, all of which were contributed by biosorption, without enzymatic
biodegradation. Live cultures of T. asperellum LBKURCC1 were not found to
bioremediate AB25 dye at pH 5.5 and 6.5. From the results of the analysis, it can
be concluded that live cultures of T. asperellum LBKURCC1 were not able to
enzymatically biodegrade BB66 dye at pH 6.5 and AB25 dye at pH 5.5 and 6.5.
Crude extract laccase of T. asperellum LBKURCC1 with enzyme activity of 0.007
U/mL and 0.021 U/ml per reaction was not effective for degrading dye BB66 and
dye AB25, with and without mediator 1-hydroxybenzotriazole hydrate (HBT), at
pH 5.5 up to six days.
