Trnl-Trnl-Trnf Intergenic Spacer Tumbuhan Rimbang (Solanum Torvum Sw.)
Rimbang (Solanum torvum Sw.) is a species of the eggplant tribe. There are many shrubs in tropical climates. DNA barcoding is one of the technological advances in the field of genetics which was developed to identify all organisms using short pieces of DNA. One of the DNA barcodes used in molecular analysis is trnL-trnL-trnF Intergenic Spacer, because it has a high mutation rate and variability. The aim of this study was to analyze the DNA barcode sequence in the trnL-trnL-trnF IGS of rimbang (Solanum torvum Sw.) taken in the of Pekanbaru city, Riau province. The methods of this research are total DNA isolation, total DNA electrophoresis, PCR, electrophoresis of PCR products, sequencing and data analysis using BioEdit, BLASTn, and MEGA6 software. The results showed that the DNA sequence of trnL-trnL-trnF Intergenic Spacer obtained had a size of 939 bp and had the highest similarity with Solanum torvum (99,79%). The results of the study showed that there were 10 nucleotide variations and 2 critical nucleotides. The Critical nucleotides are the nucleotides that could differentiate between the rimbang and the accessions studied. Rimbang formed a group with Solanum torvum and Solanum lanceolatum.
