Optimalisasi pH, WAKTU, Dan Suhu Produksi Enzim Selulase Dari Jamur Endofitik Fusarium oxysporum LBKURCC41
SUMMARY
Cellulases are enzymes that catalyze the hydrolysis of cellulose, they are produced by fungi and bacteria. Fusarium oxysporum LBKURCC41 were isolated from the tubers of Dahlia plants (Padang Luar, Sumatera Barat) and identified as endophytic fungi which showed ability to produce antimicrobial agent saponin and enzymes of β-galaktosidase and inulinase on studies before. This study aims to determine the potential of endophytic fungi Fusarium oxysporum LBKURCC41 to produce cellulase enzyme, and proceed by determine the optimum culture conditions of pH, incubation time, and temperature to yield the maximum cellulase activity. The enzyme activity obtained based on the amount of reducing sugar concentration in the crude extracts which assayed by the 3,5-dinitrosalicylic acid (DNS) method. The specific activity of enzyme was determined by estimating the protein concentration in the crude extracts using the Lowry method. The result showed that the fungi can produce cellulase enzyme and the optimum culture condition obtained at pH 5,5, incubation time 7 days, and temperature at 30°C. The maximum enzyme activity and spesific activity were 125,04 ± 0,02 x 10-3 U/ml and 447,80 ± 0,06 x 10-3 U/mg respectively.
