Induksi Kalus Dari Kotiledon Jeruk Kasturi (Citrus Microcarpa (Bunge) Wijnands) Dengan Penambahan Beberapa Kombinasi Zat Pengatur Tumbuh Secara In Vitro
Kasturi orange (Citrus microcarpa (Bunge) Wijnands) is a medicinal plant that contains chemical compounds from secondary metabolites. The content of secondary metabolites can be produced quickly using in vitro callus induction. This study aimed to determine the effect of the combination of 2,4 D, BAP, and GA and determine the effective concentration of a combination of 2,4 D, BAP, and GA on callus induction of orange cotyledon explants using in vitro method. This study used Complete Randomized Design (CRD) which consisted of seven treatments (control, 1 mg/L 2,4 D + 3 mg/L BAP, 3 mg/L 2,4 D + 3 mg/L BAP, 5 mg/L 2,4 D + 3 mg/L BAP, 1 mg/L GA + 3 mg/L BAP, 3 mg/L GA + 3 mg/L BAP, and 5 mg/L GA + 3 mg/L BAP) with four replications for each treatment. Data obtained from observations were analyzed using ANOVA. If there was a significant effect, DMRT test was then carried out at the level of 5% using the application version 17.0. Data from callus morphology were analized descriptively. The results showed that treatment of a combination of 2,4 D, BAP, and GA had no effect on callus induction of orange cotyledon explants. The treatment of 5 mg/L 2.4 D + 3 mg/L BAP produced the highest callus growth (+++) and the fastest callus time was 5.25 DAP and 1 mg/L 2.4 D + 3 mg/L BAP produced the highest percentage of callus formation 87.50%. The combination of GA and BAP produced callus, as well as shoots and roots.
