Hemybagruss nemurus is one of substantial edible fishes in Indonesia.
Hemybagruss nemurus has a soft and white thick flesh without any smooth bones
that made this kind of fish is popular in society especially for people in Sumatra
such as Jambi and Riau. The high demand of Hemybagruss nemurus has caused
the fishing intensity of this fish in public waters increased, which resulted the
population of Hemybagruss nemurus’ in its natural habitat are decreases. As one
of efforts to cope the people demand over Hemybagruss nemurus is to do
cultivation activities. The cultivation of Hemybagruss nemurus is similar with
other fishes which started with hatchery. Currently, the challenge in hatchering of
Hemybagruss nemurus is there still not found yet any optimum diluent solution for
spawning this fish. As for one tool to help increasing production in spawning this
fish is by using physiological solution. Physiological solution is often used in
hatchery process as a solution for diluent the fish sperm. However, the use of
physiological solution that are commonly sold in the market still produced a low
quantity of spawning. In the artificial spawning process, the temperatures have
an important role in fertilization process. So that it is important to provide
proper and good temperature during fertlization. Therefore, it is important to
conduct a study to find out the optimum temperature and NaCl solution dose also
the effect they gave to survival rate and the time needed for hatching
Hemybagruss nemurus’ egg. This study aims to find out the effectiveness of NaCl
and different temperatures during fertilization on the value of fertilization,
hatchability of eggs and survival rate of Hemybagruss nemurus larvae.
This study was conducted in March – April 2020 at Balai Benih ikan
(Fish seed hall) of agriculture faculty of Islamic University of Riau (UIR)
laboratory and Fish Hatchery and Breeding laboratory, Faculty of Fisheries
University of Riau (UNRI) in Pekanbaru city, Riau province. this study used
experiment method using RAL factorial with two factors. The first factorial is
concentration of different NaCl diluent solution consisting of 0,3% ,0,5% , 0,7%
and 0,9 % dose. The second factorial is the different temperatures during
fertilization consisting of 100C , 150C, 200C dan 250C temperatures. To avoid the
deviation, each treatment was repeated in three times.
The research procedurein this study using the broodstock of male and
female Hemybagruss nemurus fish which were used with an average age of 1.5 - 2
years and already mature Gonad with Gonad maturity level (TKG IV). The
amount of fish being spawned were 10 male fish and 5 female fish. To stimulate
the ovulation process, the male and female broodstock fish were injected by using
stimulating substance ovaprim. The injection dose for the female broodstock is 1
ml kg / body weight and the male is 0.5 ml / kg body weight, the injection process

is carried out once with a latent time of approximately 15 hours. The diluent
solution made for each treatment was as much as 500ml with different solution
concentrate of 0,3%, 0,5%, 0,7% dan 0,9% dose. The basic ingredient of the NaCl
solution is an infusion solution with 0.9% Nacl content and aqudes as a diluent.
To adjust the temperature of the diluent according to the treatment are in 100c,
150c, 200c and 250c temperatures which made in such a way that the container
with the coolant is ice cubes / krosok ice. The container being used is sterofoam
box size 30cm x 50cm x 30cm. This box was divided into two bulkheads, first
bulkheads was filled with ice cubes as high as 15cm while the second bulkheads
was not filled with ice cubes. To make the 100c temperature, the solution that has
poured into aqua bottle size 600ml was put into the ice cubes as deep 12-13cm
with the bottom of bottle as deep as 3cm. While for making the 150c temperature,
the solution in the aqua bottle was put on the pile of ice cubes. For the 200c
temperature, the solution in the aqua bottle was put in the bulkheads which has no
ice cube then the sterofoam was closed tightly. Each bottle was put thermometer
which has monitor on the tip of the cable, so that it can be seen from the outside.
For the 250c temperature which same as room temperature, it was put on the
sterofoam and then being left for about 15 minutes so that each desired
temperature measurement is constant. After that the study was conducted to
observe fertilization and also to measure the motility of cement level of the fish by
using microscope. The fish sperm of Hemybagruss nemurus were obtained from
the surgery of the fish by using scalpel for then being taken its Gonad and
destroyed. After that the fish sperm being destroyed was filtered using a small dip
then the cement liquid is collected using a small bowl then it is put into the diluent
solution according to the treatment dose.the fish egg was obtained by doing
stripping on the female broodstock and was collected using bowls. The fish egg
and sperm were combined according to the diluent solution then stirred with the
chicken feathers. The fertilized eggs are then incubated in the hatching medium.
Water quality parameters observed included: temperature, pH value, and
dissolved oxygen (DO) content. Dissolved oxygen content is measured using a DO
meter. Water temperature is measured using a thermometer (oC) and pH value is
measured with a pH meter (numbers 1-14). Motility data and duration of
spermatozoa motile were analyzed descriptively, meanwhile, the number of eggs,
the degree of fertilization and hatching and the survival of larvae aged 3 to 7 days
were analyzed by ANOVA test (Sudjana, 1991). By using SPSS version 22.0
software. If there is a significant difference (95%), then proceed with the
Newman-Keuls range test.
The results of this study show that the best treatment was a diluent
solution with a concentration of 0.5% NaCl and a temperature of 15oC. The
results obtained were spermatozoa motility is 4 and motility duration is 43.67
minutes, fertilization value is 97.1%, hatching value is 94%, survival rate of 3
days is 97.5%, larva survival rate of 7 days is 99%, and abnormal larvae value is
1,6%. The results of this study suggested that the Hemybagruss nemurus fish
spawning process requires accuracy and speed in conducting the fertilization, so
that, the ability of sperm to fertilize Hemybagruss nemurus fish eggs can produce
optimum fertilization. In addition, the accuracy in measuring NaCl and
temperature requires an accurate measuring device so that the fertilization

process does not experiencing any problems because the time in the fertilization
process is very short. Based on the results of this study, it is hoped that in the
future the sustainability of this research can be applied to other types of fish.
