Uji Antagonis Beberapa Isolat Bakteri Endofit Terhadap Xanthomonas Sp. Dan Karakterisasi Fisiologis Biokimianya Secara In Vitro
ABSTRACT
This study aims to obtain biochemical physiological characters and to test endophytic bacteria that have antagonistic activity against Xanthomonas sp. This research was conducted with experimental methods and observation methods. Endophytic bacterial antagonist test against Xanthomonas sp. in vitro with experimental methods was carried out using a completely randomized design (CRD) with 21 treatments which were isolates of endophytic bacteria from oil palm, acacia and eucalyptus (B) plants which were repeated three times, hypersensitivity test on endophytic bacteria by observation method on tobacco leaves, physiological biochemical characterization of endophytic bacteria by observation method. The parameters observed were the antagonistic power of endophytic bacteria isolates against Xanthomonas sp. in vitro, hypersensitivity test, physiological biochemical characterization of endophytic bacterial isolates. The results showed that 20 isolates of endophytic bacteria had antagonists against Xanthomonas sp. Eleven isolates of endophytic bacteria had higher inhibitory power, namely D1, 066B, P3, A1, 068B, B2, 067B, B1, B3, P2, 062B against Xanthomonas sp. in vitro. The results of the hypersensitivity test showed that the endophytic bacteria tested did not show necrotizing symptoms on tobacco leaves. Eleven endophytic bacteria showed positive reactions in the catalase test, oxidase test, of test and starch hydrolysis and were suspected to have similarities with the genus Bacillus bacteria based on the results of their physiological and biochemical characterization.
