Pengaruh Pemberian Hormon Rgh Terhadap Aktivitas Enzim Protease Dan Rasio Rna/Dna Ikan Patin Siam (Pangasianodon Hypophthalmus) Yang Dipelihara Pada Media Bersalinitas
Several freshwater fish commodities have been developed to adapt to
saline media, including the Siamese catfish (Pangasianodon hypophthalmus). The
growth of is fish is relatively slow, it takes around 3-8 months to harvest. To
overcome this problem, growth hormone is given in the form of Recombinant
Growth Hormone (rGh), as it is reported to have various effects, including
increasing protein metabolism. Protein derived from feed cannot be directly
absorbed by the intestine, therefore the protease enzyme acts as a catalyst. The
physical appearance or phenotype of an organism is the result of metabolic
processes that occur in every cell that makes up the organism.
The purpose of this study is to analyse the effect of rGh hormone on
protease enzyme activity, RNA/DNA ratio and growth of catfish reared in saline
media. A CRD with 4 treatments and 3 repetitions was applied. The treatments
applied were the hormone dosages, namely rGh at a dose of 0, 1, 2 and 3 mg/kg
feed. The fish used in this study were 8.7 cm TL of P.hypophthalmus fingerlings.
The fishes were reared in saline media (6 ppt) for a 60 days period. Parameters
observed in this study were protease enzyme activity, RNA/DNA ratio, absolute
weight growth, specific growth rate, feed efficiency and water quality.
Based on observations during the study, the best protease enzyme activity
(U/minute) and ratio of RNA/DNA were obtained in the hormone dose of 3 mg/kg
feed with respective values of 0.75 ± 0.24 U/minute and 3.30 ± 0.69. While
Absolute Weight Growth (g), Specific Growth Rate (%) and Feed Efficiency (%)
the best dose is 2 mg/kg, they were 9.23 ± 1.83 g, 15.39 ± 3.06% and 40.45 ±
0.82% respectively. Water quality values during the study were as follows:
temperature 25-29°C, pH 4.36-8.2, DO 3.3-5.5 mg/l and total ammonia 0.56-2.50
mg/l.
