Pengaruh Iaa Dan Bap Terhadap Pertumbuhan Embriosomatik Fase Globular Pisang Kepok Tanjung (Musa Paradisiaca L.) Secara In Vitro
This study aims to determine the interaction between IAA and BAP on the growth
of embryosomatic globular phase of banana kepok tanjung and to identify the
optimal combination of IAA and BAP to support the growth of embryosomatic
explants in the globular phase of banana kepok tanjung. The research employed
a factorial experiment design, arranged in a randomized complete block design
with two factors: IAA (0; 0.5; 1 and 1.5 mg.l-1) and BAP (0; 2; 4 and 6 mg.l-1).
The parameters observed included shoot emergence, number of shoots, shoot
height and percentage of successful shoot formation. Data were analyzed using
analysis of variance (ANOVA) and further tested using Duncan's New Multiple
Range Test (DNMRT) at a 5% significant level. The results demonstrated that the
interaction between IAA and BAP significantly affected bud emergence time,
shoot height and root length. IAA application influenced bud emergence time,
number of buds, shoot height and root length, while BAP application influenced
bud emergence, number of buds, shoot height and root length. The combination of
0,5 mg.l-1 IAA and 4 mg.l-1 BAP resulted in the fastest shoot emergence
(8,43 hours after treatment) and the highest shoot length (4,97 cm), whereas the
combination of 1 mg.l-1
IAA and 4 mg.l-1 BAP produced the highest number of
shoots (4,50 shoots) and the longest roots (3,93 cm). The percentage of successful
shoot formation ranged from 77,80 – 100 %, attributed to contamination and
browning.
