Isolasi Gen Aktin Pada Pandan (Pandanus Sp.) Dari Danau Kajuik Kabupaten Pelalawan Provinsi Riau
Pandanus sp. is one of the plants that are often exposed to adverse environments and has adapted well to the floodplain in Kajuik Lake Langgam, Pelalawan, Riau Province. Information on the expression of genes involved in adaptation to environmental stress are necessary and it requires an internal controls such as actin gene. This study aims to isolate the actin gene of pandan (Pandanus sp.) from Kajuik Lake. Research procedures included leaves sampling, total DNA isolation, PCR, electrophoresis, sequencing and data analysis. Data analysis was performed using BLASTn program, MEGA Version 6.0 and Version 7.0 Bioedit. The DNA sequences obtained in this study was part of the actin gene which is about 568 bp in size. Actin-specific primer for Pandanus sp. has been designed, namely, forward 5'ATC CTT CTC CAC AGA AAA CC 3' and reverse 5'CAT ATG GCT TCT CCT CAA TT 3 '.
