Analisis Sekuens Barkode Dna Pada Daerah Trnl-Trnl-Trnf Intergenic Spacer Pada Tumbuhan Terung Asam (Solanum Ferox L.)
Sour eggplant (Solanum ferox L.) is a thorny shrub from the Solanaceae
family which is generally distributed in the tropics. DNA barcodes are short
sequences that can be used in DNA barcoding techniques. One of the DNA
barcodes in plants is the trnL_trnL_trnF intergenic spacer sequence which has
high variability. The purpose of this study was to analyze and sequence DNA
barcodes in the trnL_trnL_trnF intergenic spacer region of sour eggplant
(Solanum ferox L). Samples taken from the district of Kampar, Riau as many as 3
individuals. The stages of the research carried out were survey sampling locations,
sampling, morphological observations, total DNA isolation using the GeneAid
Mini Plant kit, total DNA electrophoresis, PCR using the DreamTaq DNA
polymerase (Thermo Scientific) kit, electrophoresis of PCR products, sequencing
and data analysis using BioEdit, BLASTn and MEGA software version 6.0. The
results of morphological observations showed that sour eggplant has special
characteristics, namely the fruit is buni type, has feathers but is easy to clean, and
when it is not ripe it is green and when it is ripe it is yellow. The sequence length
obtained in the 3 samples has a length of 1,060 bp. In this study, there was no
identity value that reached 100%, the highest identity value was only 99.81%.
There were 13 nucleotide variations in this study, namely, at positions number 80,
95, 107, 243, 718, 727, 806, 1,034, 1,048 which were caused by substitution
mutations. This trnL-trnL-trnF intergenic spacer DNA sequence was the first to
be reported in Solanum ferox L.
