Studi Anatomi Dan Histokimia Daun Ruku-Ruku (Ocimum Tenuiflorum L.)

Ruku-ruku (Ocimum tenuiflorum) is one species belongs to Lamiaceae family that has a shrub habitus with an average height of 30-60 cm and single-leafed. Ruku- ruku leaves have medicinal properties. This study aims to characterize the anatomical structure of ruku-ruku leaves and determine the location of secondary metabolite compounds contained therein. The methods used were paraffin, paradermal, and histochemical methods. The anatomical structures observed include the mother leaf blade, intercostal, leaf margin, and petiole. The results of anatomical observations show the characterization of each tissue, namely epidermis, basic tissue, transport bundles and trichomes. The leaf blade has collateral-type transport bundles with additional smaller transport bundles on the adaxial side. The petiole is kidney-shaped with collateral-type transport bundles. Stomata are diasitic type with the highest stomatal density on the abaxial side 116, 67 ± 7.22/mm². Non-glandular trichomes are found on both sides of the leaf, the adaxial part of the non-glandular trichome density value of 12.5 ± 0.00/mm² is higher than the abaxial leaf density value of 9.58 ± 5.05 /mm². Abaxial surface glandular trichome density was 12.5 ± 0.00/mm². Histochemical test results show that the presence of phenol compounds, alkaloids, and flavonoids are found in all parts of the leaf. Tannin compounds are found in the mother leaf bone and intercostal, while lipid compounds show lower compound content because they are only found in the cuticle and trichomes.
