Skrining, Karakterisasi Dan Potensi Bakteri Selulolitik Asal Tandan Kosong Kelapa Sawit (Elaeis Guineensis Jacq.) Di Lahan Gambut
Empty fruit bunch of oil palm (EFBP) is hard to decomposed because the content of the constituent that is 45.9% of cellulose, 46.5% of hemicellulose, and 22.8% of lignin. One of the microorganisms which is capable of cellulose degrading is cellulolytic bacteria. Cellulolytic bacteria are able to degrade cellulose to produce cellulase enzymes. This study aims to selecting and characterizing the potential cellulolytic bacterial of EFBP in peatlands in order to obtain cellulolytic bacteria that degrade the organic matter and can be grown on acid soils. The method used is the method of explorative. EFBP samples taken at Sei Mandau village, Siak district. The isolation of bacteria carried by pour plate and scratch plate method on Nutrient Agar media and Carboxymethil Cellulose. The observation and identification conducted by macroscopic, microscopic, through the series of biochemical tests and cellulolytic bacteria potential test that is cellulolitic index, low pH test, optimization of growth and reducing sugar test. The results obtained five isolates that bacteria genus, Aeromonas, Cellvibrio, Vibrio, Serratia and Streptococcus. Cellulolytic bacteria had a potential to decipher the cellulose which can be seen from the cellulolitic index value, there are 4 isolates that have cellulolytic index ≥ 2.5, bacteria isolates with the highest cellulolitic index value is TKKS7 with the value IAS 7. Which is obtained two isolates of bacteria that can grow at pH 4 is TKKS1 and TKKS7. The optimum time of bacterial TKKS1 and TKKS7 growth are on the second day.
