Analisis Keanekaragaman Genetik Tiga Populasi Matoa (Pometia pinnata Forst & Forst) Berdasarkan Penanda RAPD (Random Amplified Polymorphic DNA)
Matoa is one of the plants originating from the Sapindaceae family which is spread in tropical areas, such as Indonesia. Matoa is a delicious fruit to consume with a distinctive taste from a combination of flavors, longan, rambutan and durian. Riau has a population of yellow, red and green skin matoa. Although these types can be distinguished morphologically, there is no information whether these populations are genetically different or not. To answer this question, it is necessary to analyze the genetic variation of tomatoes using random aplified polymorphic DNA (RAPD) markers. This study aims to determine the genetic diversity of three matoa populations based on RAPD markers.
This research was conducted from February 2023 to April 2023 at the Genetics and Molecular Biology Laboratory, Faculty of Mathematics and Natural Sciences, University of Riau. Matoa samples were taken from Kampar Regency (Tapung Hilir District), Pekanbaru City (Rumbai and BBI Pekanbaru), Riau Province. The samples used in this study were three matoa populations. Each population is occupied by 5 individuals. Total DNA isolation from matoa plants was carried out using the Genomic DNA Mini Kit Plant (Geneaid). The primers used in the study included OPD-08, D-08, X-01 and OPJ-20. The results showed that the number of bands detected was 65 bands. The size of the bands produced ranged from 250 bp to 3100 bp. The percentage of polymorphic bands produced by all primers was 100%. The level of genetic diversity of matoa tested ranged from 8.748%-16.186%, which means the diversity of the three small matoa populations or their kinship is close. The results of the clustering analysis using 4 RAPD primers showed that the matoa samples tested did not group separately based on the color of the fruit skin and also the location of growth (Pekanbaru City and Kampar Regency).
