Induksi Kalus Eksplan Petiol Pasak Bumi (Eurycoma Longifolia Jack) Dengan Penambahan Bap (Benzyl Amino Purine) Dan 2,4-D (Dichloropenoxyacetic Acid) Secara In Vitro
Pasak bumi (Eurycoma longifolia Jack) is a medical plant belonging to the
Simaroubaceae family. This plant is rare to find due to several factors, such as
illegal logging, the location of the stamen is lower than the location of stigma that
makes the pollination process tough, the low percentage of seed germination in
natural habitats. Therefore, it is necessary to do alternative propagation of pasak
bumi in a fast time. The aims of this research are to determine the effect of giving
BAP, 2,4-D alone and the interaction of the two in callus formation and to determine
the best concentration of BAP, 2,4-D and the interaction to induce callus explant
petiole Eurycoma longifolia Jack in vitro. This research was conducted at the
Terpadu Laboratory, Departement of Biology, Faculty of Mathematics and Natural
Science, the University of Riau from April-June 2021 using RALF with 2 factors.
The first factor is BAP with 4 concentration levels, are 0 ppm, 0.5 ppm, 1 ppm, 1.5
ppm. The second factor is 2,4-D with 4 levels of concentration, are 0 ppm, 2 ppm,
4 ppm, 6 ppm. The results of this research showed that the concentration of BAP
and 2,4-D significantly interacted with the time of callus emergence. The
combination of 0.5 ppm BAP + 2 ppm 2,4-D was the best treatment with the fastest
callus emergence 7 day after planting with the percentage of live explants and the
percentage of callus being 100% each, the highest callus growth (++++) with 100%
crumb callus texture.
