Fraksinasi Ekstrak Polar Senyawa Antioksidan Kulit Manggis (Garcinia Mangostana Linn) Menggunakan Kromatografi Gel Filtrasi
Garcinia mangostana fruit hull contain antioxidant compounds. A
concentrated extract of Garcinia mangostana fruit hull in 50% ethanol and
0.05M NaOAc pH 5,5 buffer, had an in vitro antioxidant activity (IC50) of
47.3 μg/mL, when measured by the 2,2-Diphenyl-1-picrylhydrazyl (DPPH)
method. However, this concentrated extract was toxic when applied to
keratinocyte cells, with IC50 of μg/mL. This was due to the high content of
salt in the concentrated extracts. To separate the antioxidant activity from
the smaller molecular weight salts contained in the extract, the extract was
fractionated by the gel filtration column chromatography method, using
Sephadex LH-20. The column was eluted with methanol, and each 25
mL of the eluted filtrate collected. The fractions were then concentrated by
vacuum rotary evaporation and dried to constant weight at 50oC. Three
fractions gave higher in vitro DPPH antioxidant activity than before
fractionation. Fraction 2, 3 and 7 had IC50 of 12.8 μg/mL, 19.3 μg/mL and
10.5 μg/mL respectively.
