Optimalisasi Produksi Senyawa Antimikrob Jamur Endofit Sporothrix Sp. (Lbkurcc43) Menggunakan Sumber Karbon Dan Nitrogen Dari Bahan Alam
Endophytes are organisms living in plants tissue which can produce antimicrobial compounds as the host. Production of chemical compounds by fermentation is influences by media composition. Fermentation media is the main component to produce industrial compounds. The objective of this research was to investigate optimal media consisted of widely available natural compounds in order to produce antimicrobial compounds from endophytic fungus Sporothrix sp. LBKURCC43. The media were modified by applying composition combination between corn, potato, and sweet potato as carbon sources, as well as soybean and beef meat as nitrogen sources at incubation times of 5, 10, 15, 20, and 25 days. Antibacterial activity against pathogenic microorganisms (Escherichia coli, Staphylococcus aureus, and Candida albicans) was examined by using agar diffusion method. The media with the highest activity determined by phytochemical test. The compounds produced in modified medium MM 03 (potato and soybean) showed the highest inhibition zone diameter (8.09 ± 0.62 mm) at incubation day 20 against E.coli, with the ratio between potato and soybean was 3.60 : 0.24. Meanwhile, the compound in medium MM 05 (corn, potato and soybean at ratio of 1.8 : 1.8 : 0.24) indicated the highest inhibition zone against S. aureus (6.03 ± 0.82 mm) at day 20. However, none of the compound in modified medium was active against C. albicans. In conclusion, the ability of endophytic fungus in producing antimicrobial compounds was determined by the used medium composition. The highest activity was obtained in the media with composition of potato and soybean on the E.coli patogen. Phytochemical test indicated that modified medium MM 05 contained compound categorized into saphonin.
