SUMMARY
Pulai basung (Alstonia spatulata Bl) is a type of tree that is widely spreaded throughout Indonesia with medicinal content in it. This plant belongs to the genus Alstonia and the family Apocynaceae. This plant is a tree that usually lives in swamps with grayish smooth skin and yellow inner skin and has abundant sap. The purpose of this study was to isolate and analyze the spectroscopy of secondary metabolite compounds of the dichloromethane fraction pH 9 and analyze the toxicity test of the chloroform fraction pH 8 of the stem bark of the
A.spatulata plant using the BSLT method. This research uses acid-base extraction method. Samples were extracted by maceration method using methanol solvent and partitioned using chloroform (pH 8) and dichloromethane (pH 9). The results of the KLT chromatogram of the two fractions show the number of stains found in both extracts with the same shape. In this study, the isolation process continued using the dichloromethane fraction pH 9 with separation using preparative KLT while the chloroform fraction pH 8 was continued for toxicity testing. The results of preparative KLT produced 5 bands followed by UV-Vis spectroscopic analysis with maximum absorption at a wavelength of 247.0 nm while the FTIR spectrophotometer showed the impure compound ASP-D-PRE-P1 had a wave number of 3079, 49 cm-1 (C-H aromatik), 2958 cm-1, 2925 cm-1, 2873 cm-1, 2851 cm-1 (C-H aliphatic), 1603 cm-1 (C=C aromatik), 1358 cm-1 (C-N), and 1261 cm-1, 1239 cm-1, 1209 cm-1 (C-O). Toxicity test results using the BSLT method showed that the chloroform fraction pH 8 was toxic with an LC50 value of 292,32 ppm.
