Pengaruh Ukuran Eksplan Daun Dan Kombinasi Bap Dengan Naa Terhadap Perkembangan Eksplan Jaringan Daun Sansevieria (Sansevieria Macrophylla)
Sansevieria macrophylla is one of the ornamental plants that has the economic potential to be cultived in Indonesia. Applying in-vitro culture to the sansevieria plants propogation which is enables the production of several superior new plants in a short amount of time. In-vitro propogation of Sansevieria uses explant from different size explant, along with addition of BAP and NAA to the culture medium. The objectives of this study were to determine the effect and find the best explant leaf, combination of BAP + NAA and the combination of explant leaf with BAP + NAA on development of Sansevieria macrophylla leaf tissues explants. The research was conducted at plant Biotechnology Laboratory, Faculty of Agriculture, University of Riau, from December 2022 to July 2023.The study used a factorial experiment with completely randomized design (CRD) consisting 2 factors with 16 combinations and 3 replications. The result showed that the interaction of different size with BAP + NAA, different size explant as a single factor and treatment of BAP and NAA as a single has effect to the explant Sansevieria macrophylla development. The research results showed that the interaction of different explant size and the combination of BAP + NAA on MS media, the single factor of explant size, and the single factor of the combination of BAP + NAA on MS media has effect on callus appeared, callus weight, when shoots appeared, and the number of shoots on Sansevieria macrophylla leaf tissue explants. The best explant development was produced by the combination of size explant 1 cm x 1 cm x 0,5 cm with BAP 0 mg.l-1 + NAA 2 mg.l-1 on the time of callus emerged which is 31,23 DAP, weight reaches 4,86 g, the number of shoots per explant is 2 shoots and the number of planlets per explant is 2 planlets.
