Eksplorasi Dan Uji Antagonis Bakteri Rizosfer Asal Tanaman Kelapa Sawit Terhadap Ganoderma Sp. Secara In-Vitro
ABSTRACT
This study aims to explore and test the antagonistic power of rhizosphere bacteria from oil palm plants which can control Ganoderma sp. in vitro causes of oil palm stem rot disease. This study was carried out at the Plant Disease Laboratory, Faculty of Agriculture, Riau University. This study was carried out from June to October 2023. This research was conducted using a qualitative descriptive method consisting of 3 stages, namely: 1) exploration including isolation and purification of rhizosphere bacteria from oil palm plants, 2) characterization of rhizosphere bacteria from oil palm plants including morphological characterization and physiological characterization, and 3) test the antagonistic power of rhizosphere bacteria from oil palm plants against Ganoderma sp. in vitro. The data obtained was analyzed descriptively in the form of tables and figures. The results showed that the rhizosphere bacteria from oil palm plants (A1.1, A1.2, A2.3, A2.5, B1.2, and D2.2) that were isolated showed morphological characteristics of white color, round or circular shape, elevation flat, and has irregular edges. The results of observations of the physiological characteristics of rhizosphere bacteria from oil palm plants during the catalase test and oxidase test reacted positively except isolate A1.2 showed a negative reaction in the catalase test, and isolate B1.2 showed a negative reaction in the oxidase test. All bacterial isolates were oxidative during the oxidase-fermentation test and were Gram positive bacteria. The results of the antagonist test for rhizosphere bacteria from oil palm isolates A1.1 and A1.2 were able to inhibit the growth of Ganoderma sp. isolate LPTUNRI-Gan003 with an average clear zone index A1.1 of 1.12 and A1.2 of 1.41.
