Isolasi Bakteri Escherichia Coli Dan Vibrio Sp Pada Tambak Udang Vaname (Litopenaeus Vannamei)
Escherichia coli is a common indicator organism used to assess water quality, particularly for fecal contamination. The presence of E. coli in vannamei shrimp ponds can indicate contamination from human or animal waste, which can lead to decreased water quality and health risks for consumers of fishery products. Meanwhile, Vibrio sp is a group of pathogenic bacteria that can cause various diseases in shrimp, including Taura syndrome and white syndrome.This research was conducted from March to May 2024. The isolation and selection of Escherichia coli and Vibrio sp were carried out at the Marine Microbiology Laboratory of the Department of Oceanography, Faculty of Fisheries and Marine Science. Samples were taken from vannamei shrimp farms in Ketaping Village, Padang Pariaman Regency, West Sumatra. This study aims to isolate and identify E. coli and Vibrio sp present in shrimp, water, wastewater treatment plants, and sediments cultivated in shrimp farms in Ketaping Village, Padang Pariaman Regency, West Sumatera.The method used in this research is the survey method, and for sample handling, isolation was performed, followed by isolate characterization and biochemical tests. In this study, the results of the coliform bacteria abundance estimation test showed the highest count in the wastewater treatment plant, with ≥1898 MPN/100 ml. The confirmatory test results revealed the highest fecal coliform bacteria abundance in four samples: effluent water, pond water, sediment, and vannamei shrimp, with a value of ≥1898 MPN/100 ml. Further confirmatory tests were conducted to determine the presence of E. coli bacteria in the samples. Out of the 6 samples tested, 5 were found to be positive for E. coli bacteria, including sediment samples (SDC2, SDC3), shrimp samples (UDA2, UDC3), wastewater treatment plant samples (IPA3, IPC1), influent water (AMC2), and effluent water. (AKA2, AKB2, AKA3). The isolation and identificationiresults were obtained: shrimp isolate (UD2), sediment (SD1, SD2, SD3), wastewater treatment plant (IP1), inlet water (AM1, AM2), oulet water (AK1, AK2), and pond water (AT1, AT2).
