Studi Filogenetik Genus Mangifera Di Sumatera Bagian Selatan Berdasarkan Sekuen Dna Daerah Its
The diversity of Mangifera in South of Sumatera has not been explored and studied. On the other hand, the diversity of Mangifera. is endangered due to the lost of its natural habitat. These factors may result less information of Mangifera, whereas the diversity and taxonomical status are not yet clear. The aim of this study was to reconstruct the relationship of Mangifera based on the internal transcribed spacer (ITS) region of nuclear ribosomal DNA (nrDNA). DNA was isolated from leaves using CTAB method with a modification. DNA sequencing was done in First Laboratories, Malaysia. The phylogenetic reconstruction was carried out using PAUP* Version 4.0b10, Maximum Parsimony (MP) and Neighbor Joining (NJ). Cladogram using MP showed monophyletic tree and devided into two main clades. The first clad consisted of M. foetida1, M. foetida2, M. foetida3, M. odorata, M. kemanga1, M. indica1, M. indica2, M. zeilanica and M. lalijiwa and the second clad consisted of M. torquenda, M. quadrifida and M. kemanga2. The NJ cladogram supported MP analyses and showed that M.indica2 has the longest genetic distance to other species.
