Isolasi, Identifikasi Morfologi, Dan Uji Aktivitas Enzim Hidrolitik Dari Bakteri Termofilik Sumber Air Panas Pawan Rokan Hulu, Riau
Thermophilic bacteria is microbe that can grow at temperatures of 45-80°C and potential in producing enzymes stable to heat. Thermostable enzymes have a number of advantages in their use in the fields of industry and the world of biotechnology. The samples were taken from hot springs in the Pawan area, Rokan Hulu Regency (Rohul), Riau Province. This study uses duplicate repetition of isolation pour plate method with three variations in incubation temperature, namely 45°C, 50°C, and 55°C. The morphology identification of isolates was carried out macroscopically and microscopically to determine the genus of bacterial isolates accompanied by biochemical tests. The ability test of bacterial activity in producing thermophilic enzymes was carried out using the disc diffusion method. Based on the results, 17 isolates were obtained with three incubation temperature variations, namely 6 isolates at 45°C (LBKURCC229-LBKURCC234), 6 isolates at 50°C (LBKURCC179-LBKURCC184) and 5 isolates at 55°C (LBKURCC255-LBKURCC259). Seventeen of these isolates have the genus of Bacillus, Pseudomonas and Thermus. The activity of hydrolytic enzymes is known by the presence of clear zones formed on selective media for starch, inulin, CMC and lipids. The ratio of clear zones for each enzyme activity was obtained: for amylase enzyme 6 isolates with a ratio value not significantly different P≤0,05 with a ratio range of 2,07-2,44: that is LBKURCC 231 (Pseudomonas) at 45°C, and LBKURCC 179, LBKURCC 181-184 (Bacillus), at 50°C. The clear zone ratio for cellulase is LBKURCC 256 (Bacillus) of 3.93 ± 0.37 at 55°C with a high category. Clear zone ratio for inulinase namely LBKURCC 231 genus Pseudomonas 3,03 ± 0,02 (high category). Whereas all isolates did not produce clear zones on lipid agar media. Based on data from the results of this study indicate that isolates that have the potential to produce hydrolytic enzymes are isolates with high clear zone ratio criteria.
