Perbandingan Uji Slide Aglutinasi Dengan Uji Polymarase Chain Reaction (PCR) Untuk Mendiagnosa Penyakit Enteric Septicemia Of Catfish (ESC) Pada Ikan Patin (Pangasius Pangasius)
Agglutination occurs due to antibody and antigen reaction may be used as a basis to develop a test kit to detect the presence of bacteria that cause fish disease rapidly. To understand whether the antibody- antigen reaction of E. ictaluri can be used to detect the Enteric Septicemia of Catfish (ESC) disease, a research has been conducted from September to December 2018. Antibody was collected from rabbits that was immunized with inactive E. ictaluri (multilevel doses, 4 weeks period, 0.5; 1 ; 2; and 3 ml/week respectively). Serum antibody was harvested in the 5th week. The antibody was then used to detect the presence of E. ictaluri in the infected fish. Ten Pangasius pangasius were infected with active E. ictaluri (1 ml, 106CFU/ fish, intraperitoneal injection). Three days after the infection and fish showing the ESC symptom, the E. ictaluri isolate was taken by extracting liver and kidney and then the isolate was then grown in the TSA media for 24 hours. The E. ictaluri isolate was then spread in a slide glass, and mixed with E. ictaluri antibody. The reaction of that antigen-antibody was studied. The positive reaction was indicated by the presence of granules, while negative reaction produces no granule. Results shown that the antigen of E. ictaluri collected from the ESC diseased fishes positively reacted to E. ictaluri antigen and the granules were forms within 1 minute after antigen – antibody was mixed. E. ictaluri antibody showing negative reaction toward other bacteria antigens. The results of antigen reaction and serum antibodies of E. ictaluri bacteria through slide agglutination test showed identical results with PCR test results with 98.98% squensing results and the calculation of diagnostic sensitivity and diagnostic specificity values of the test obtained 100%. Based on data obtained, it can be concluded that the Slide Agglutination Test can be used as rapid diagnostic test for detecting the ESC disease caused by E. ictaluri bacteria.
