Pemanfaatan Ekstrak Etanol Biji Mangga Harumanis (Mangifera Indica L) Untuk Menghambat Pertumbuhan Bakteri Edwardsiella Tarda
There are antibacterial compounds in the mango seed. Thus the extract of that seed may be used as antibacterial to prevent the growth of bacteria that attack fish such as Edwardsiella tarda. To understand the type of bioactive compounds present in the mango seed extract and the effectiveness of mango seed extract in hampering the growth of E. tarda, a study has been conducted in August to November 2018 in Faculty of Fisheries and Marine Sciences, and Faculty of Mathematics and Natural Sciences, University of Riau. The mango used in this study was purchased from the Pasar Pagi Arengka market, Pekanbaru. The mango seed was extracted using ethanol. The sensitivity of E. tarda toward mango seed extract was studied using a KIRBY-BAUER method. Mango extract in 19 different concentrations (200 – 10,000 ppm) were tested using 6 mm disk blanks that were placed in E. tarda cultured in TSA media, 50 µL/ disk. The clear zones formed were then measured. While the LD50 was tested using Pangasius hypopthalmus fingerlings (7-9 cm TL). Four media with different concentrations of mango extract, namely 0 ; 100 ; 200 and 300 ppm were used to keep the fingerlings (10 fingerlings/ aquarium, 3 replications) for 24 hours. The bioactive compounds of the mango seed extract were identified using a Thin Layer Chromatography method and a screening process. Results shown that E. tarda was sensitive to mango extract. The clear zone in the 200 ppm treatment was 6.44 mm, in the 300 ppm was 6.89 mm, 400 ppm was 7.11 mm, 500 ppm was 7.52 mm, 600 ppm was 7.60 mm, 700 ppm 7.92 mm, 800 ppm was 8.37 mm, 900 ppm 8.87 mm, 1000 ppm was 9.36 mm, 2000 ppm was 10.27, 3000 ppm was 10.88 mm, 4000 ppm was 11.61 mm, 5000 ppm was 12.52 mm, 6000 ppm 12.84 mm, 7000 ppm was 13.30 mm, 8000 ppm 14.12 mm), 9000 ppm 16.31 mm and 10.000 ppm was 19.07 mm.
Thin Layer Chromatography test shown that the antibacterial compounds present in the mango seed extract were tannin, flavonoid, terpenoid and saponin. The Rf of tanin was 0.78, flavonoid was 0.48, terpenoid was 0.56 and saponin was 0.44. The LD50 test shown that the lethal dose of mango seed extract toward P. hypopthalmus fingerlings was 206.56 ppm.Based on data obtained, it can be concluded that the mango extract can be used to hamper the growth of E. tarda.
