Potensi Kappa Karaginan Dari Rumput Laut (Eucheuma Cottoni) Sebagai Antioksidan Dan Inhibitor Α-Glukosidase

Seaweed is one of the primary producers in marine ecosystems including
phytoplankton, seagrass and mangroves. Seaweed has economic value as a
producer of hydrocolloids, one of which is carrageenan. Carrageenan consists of
two fractions based on their sulfate content, namely kappa carrageenan which
contains less than 28% sulfate and carrageenan iota containing more than 30%
sulfate.
The use of seaweed for the treatment of various diseases, including diseases
caused by free radicals and metabolic diseases characterized by hyperglycemia,
occurs due to insulin secretion, insulin action or both or as a chronic disease that
occurs due to the inability of the pancreas to produce sufficient insulin.
Carrageenan extracted from seaweed type Eucheuma is a phycocoloid which is a
polysaccharide compound. Eucheuma cottoni is a type of seaweed that produces
kappa carrageenan, it is known that plant cell wall polysaccharides and lignin
cannot be digested by the digestive enzymes of mammals, including humans, so
the advantages of consuming fibrous foods, especially those that are water
soluble, include reducing or inhibiting the rate of sudden increase in blood
glucose levels. Maintaining blood sugar levels alone is not enough to prevent
complications of diabetes mellitus, this must also be followed by a reduction in the
amount of free radicals in the body in excess (oxidative stress).
This study aims to (1) determine the chemical composition of E. cottonii; (2)
the physico-chemical characteristics of kappa carrageenan; (3) determine the
ability of kappa carrageenan as an antioxidant and α-glucosidase inhibitor. This
research was conducted in five stages, namely: (1) preparation and analysis of
the chemical composition of E. cottonii, (2) extraction of E. cottonii produced
kappa carrageenan E. cottonii and determined the characteristics of kappa
carrageenan E. cottonii, (3) phytochemical analysis, (4) Analysis of antioxidant
activity and (5) α-glucosidase inhibitor of kappa carrageenan E. cottonii.
The experimental design used was a completely randomized design (CRD)
with kappa carrageenan concentration treatment of K1 = 3 g, K2 = 5 g, and K3 =
10 g. The parameters observed consisted of the chemical composition of E.
cottonii, characteristics of kappa carrageenan E. cottonii, phytochemical and
antioxidant activity and α-glucosidase inhibitor. Analysis of the resulting data
used the Analysis of variance (ANOVA) and continued with the Duncan test to see
the differences in each treatment.
The results showed that the chemical composition of dry E. cottonii was
moisture content (14,23%), ash content (38,63%), protein content (5,12%), fat
(0.81%), crude fiber (22.18%), and carbohydrates (32,47%). The physical
characteristics of the carrageenan extract with 8% potassium hydroxide (KOH)
solution resulted in yield (52,00%) and viscosity (52,53 cP). Chemical
characteristics are water content (8,97%), ash (24,76%), acid insoluble ash

(0,93%), sulfate (24,50%), and the results of phytochemical analysis contained
bioactive compounds, namely saponins and terpenoids/steroids.
The highest antioxidant activity in the study was produced at a
concentration of 10 g (0.0643 mg TE/g) with percent inhibition of the α-
glucosidase enzyme (58,5085 mg/mL) compared to kappa carrageenan E. cottonii
at a concentration of 3 g and 5 g ( 0,0200 mg TE/g and 0,0515 mg TE/g) with
percent α-glucosidase inhibitor (38.6386 mg/mL and 47,4474 mg/mL). Kappa
