Efek Inhibitor Proteasom Terhadap Ekspresi Katalase Pada Aorta Tikus Aterosklerosis
Proteasome is a complex enzyme for protein degradation in most of intracellular
proteins, so that, cellular proteasomes process’s regulation plays an important
role. This research aimed to analyze the effect of giving a proteasome inhibitor,
bortezomib, on the catalase expression in the aorta of atherosclerotic rat model.
This experimental research used 15 male Wistar rats which were divided into
control group (I), atherosclerosis group (II), and atherosclerosis group by giving
bortezomib (III). Catalase expression assessed in the form of intensity and area
examined by immunohistochemical techniques, where the area shows the ratio
between the area of the aorta containing catalase and the entire aorta area, while
the intensity represents the concentration of catalase and then processed
quantitatively using Adobe Photoshop software. The results showed that the
highest average intensity of catalase expression was found in the
atherosclerosis+bortezomib group, which was 92.86 and the lowest was in the
atherosclerosis group, which was 86.20 and the highest catalase expression width
was found in the control group, which was 3.09 and the lowest was in the
atherosclerosis group, which was 1 ,63. There was an increase of catalase
expression’s intensity by giving bortezomib with an average of 92.86 and catalase
expression’s width of 2.42, statistically, however, it did not show significant
results.
